چکیده

domains, the nucleotide binding domains (NBDs), but which have evolved to couple the energy released by binding and hydrolysis of ATP to processes other than transbilayer transport. These include proteins involved in chromatin organization (13), DNA repair, telomere maintenance (15, 44), and mRNA trafficking through the nuclear pore (23). The molecular mechanism described below for ATP catalysis by the NBDs and its coupling to transport is also likely to describe the conformational changes involved in these processes.

Structural diversity of ABC transporters

ATP-binding cassette (ABC) transporters form a large superfamily of ATP-dependent protein complexes that mediate transport of a vast array of substrates across membranes. The 14 currently available structures of ABC transporters have greatly advanced insight into the transport mechanism and revealed a tremendous structural diversity. Whereas the domains that hydrolyze ATP are structurally relat...

Tracing the structural evolution of eukaryotic ATP binding cassette transporter superfamily

The ATP binding cassette (ABC) transporters superfamily is one of the largest classes of membrane proteins. The core of the ABC transporter protein is composed of transmembrane domains (TMDs) and nucleotide binding domains (NBD). Eukaryotes ABC transporters are classified into seven main families (ABCA to ABCG) based on sequence similarity and domain organizations. With different domain number ...

The ABC of channel regulation

The ABC Transporter/Channel Superfamily The ATP-binding cassette (ABC) superfamily is probably the largest and most diverse family of proteins that mediate the selective movement of solutes across biological membranes. Many ABC proteins are of considerable clinical significance: human P-glycoprotein (P-gp) confers resistance of cancers to chemotherapeutic drugs, Pghl has a role in chloroquine r...

Structure of ABC transporters.

ABC (ATP-binding cassette) transporters are primary active membrane proteins that translocate solutes (allocrites) across lipid bilayers. The prototypical ABC transporter consists of four domains: two cytoplasmic NBDs (nucleotide-binding domains) and two TMDs (transmembrane domains). The NBDs, whose primary sequence is highly conserved throughout the superfamily, bind and hydrolyse ATP to power...

ATP-binding cassette transporters are targets for the development of antibacterial vaccines and therapies.

ATP-binding cassette (ABC) transporters are widespread among living organisms and comprise one of the largest protein families. For example, components of ABC transporters are encoded by approximately 5% of the Escherichia coli and Bacillus subtilis genomes (47, 80). These transporters are found in all species and are evolutionarily related. However, they are functionally diverse and have roles...
